Originally formulated to emulate the functionality of growth hormone releasing hormones (GHRH), growth hormone releasing peptides (GHRP) took shape in an effort to replicate the actions of GHRHs. The discovery of GHRH in 1984 prompted scientists to theorize that these compounds might operate differently, with GHRPs demonstrating a noticeable affinity for ghrelin receptors. This led researchers to speculate that GHRPs, akin to the naturally occurring hormone ghrelin, could potentially modulate activities through hypothalamic stimulation, exerting influence over various physiological functions such as metabolism and nutrient absorption.

Overview

An investigation was carried out to ascertain whether the efficacy of GHRP-6 is contingent on the presence of endogenous GHRH. In a particular clinical trial, nine participants aged between 20 and 30 years were administered either a GHRH antagonist or saline, followed by GHRP-6 after a 20-minute interval. Blood samples were systematically collected over the subsequent hours, and the concentrations of growth hormones were assessed. Post-study, the findings suggested that HGH levels were diminished in subjects treated with the GHRH antagonist. The researchers theorized further that the potential action of GHRP-6 may require the presence of endogenous GHRH.

Though the intricacies of GHRP-6’s mechanism are still being explored, numerous research endeavors have indicated that GHRPs might operate by interacting with two receptors: GHS-R1a and CD36. Each subject received GHRH, GHRP-6, or a combination of both, and subsequent HGH levels were measured. Among the control group, those given both GHRH and GHRP-6 exhibited the highest HGH levels, followed by GHRP-6 alone, and lastly, GHRH alone. Conversely, subjects with hypothalamic-pituitary disconnection showed the lowest HGH levels when presented with both GHRP-6 and GHRH. This study proposed two hypotheses: firstly, GHRP-6 action may primarily occur in the presence of GHRH, and secondly, GHRP-6 may impact the hypothalamic-pituitary axis to exert its potential effects.
